Our Personalized Medicine Project is dedicated to deeply characterizing the immune mechanisms driving disease in individual patients. By accessing patients who are freshly diagnosed and have not yet received disease-specific therapies, we aim to capture an undistorted view of their immune system at a critical juncture—before therapeutic interventions alter their immune profile. To achieve this, we employ high-level proteomic and genomic analyses of patient-derived body fluids. A key component of this strategy is the whole blood activation assay, providing a functional readout of immune reactivity under near-physiological conditions. By stimulating whole blood ex vivo with defined immune triggers, we observe real-time immune responses in a complex, intact cellular environment. This approach helps us understand baseline immune activation and how patients’ immune systems respond to specific stimuli. Our ultimate goal is to identify patient-specific immune pathway signatures that can inform targeted, individualized treatment strategies, moving beyond a one-size-fits-all paradigm toward precise, mechanism-based therapy selection tailored to each patient’s unique disease biology.
