This assay provides a method for direct labeling of mRNAs and miRNAs with molecular barcodes called nCounter Reporter Probes without the use of reverse transcription or amplification. The supply arm of this resource stocks freshly prepared bacteria growth plates with or without antibiotics, transformation competent bacteria, and commercial kits and reagents for molecular biology research. On-line ordering is available and there is free delivery to the lab. The Bioanalytics Facility houses state-of–the-art instrumentation for imaging and quantitation of fluorescent, radioactive, luminescent, and chemiluminescent signals from gels, membranes, or multiple well plates. Gels and membranes stained with visible dyes or ethidium bromide can also be analyzed. Digitized images can be saved to transportable storage for further analysis in the lab or prints can be made on site. This facility also has four "real-time" PCR instruments available on a sign-out basis.

The Aperio Versa automated scanning microscope and the Leica SCN400 Slide Scanner deliver solutions for high-resolution imaging in both bright field and fluorescence. Both instruments have high-capacity robotic autoloading (200 slides for the Aperio Versa and 384 slides for the Leica SCN400) making them ideal for large slide cohorts and tissue microarrays. The associated software packages provide complex algorithms for unbiased, automated image analysis and quantitation of immunostaining in both bright field and fluorescence. In addition, these systems can be utilized as a permanent high-resolution solution for those who need archiving of histologic material. The DHSR hosts a convenient, web-based digital slide-viewing environment (Digital Image Hub) for the rapid retrieval, review, annotation and image acquisition of scanned material. This easy to use digital slide box allows researchers to share images among colleagues, both at Vanderbilt and at other institutions, from any web browser while remaining in a digitally secure environment. Built-in conferencing features allow investigators to coordinate multi-site discussions of histologic samples. Expert assistance is offered in planning experiments and processing data in a consistent, objective, and timely manner. The automated imaging and analysis performed in this core saves researchers and staff weeks of tedious work. An additional service offered by the DHSR is the creation of digital archives of critical and irreplaceable tissue samples, a benefit only feasible due to the automated high-resolution imaging of whole 25 mm x 75 mm microscope slides and 50 mm x 75 mm "double" slides. The DHSR offers specialized microscopy assistance in addition to automated slide scanning and analysis, such as colony counting, long-term live imaging and deconvolution. The core is capable of high-resolution automated and semi-automated imaging and quantitation of a wide range of cell culture and tissue samples, from bacterial colonies to oranoids.

In addition to data acquisition, we support MRI and MRS protocol development, functional MRI (fMRI) experimental design, subject preparation, structural and functional image analysis, and training on image analysis techniques and tools. Additional resources available include near-infrared spectroscopy (NIRS) and optical coherence tomography (OCT). Description The Human Imaging Core is dedicated to the application of advanced and prototypical MRI methods to study specific organ systems of the human body such as: nervous (central and peripheral), muscular-skeletal, cardio-pulmonary, and integumentary systems. Additionally, many ongoing projects focus on development and validation of novel MRI biomarkers for the assessment of numerous diseases (e.g. cancer, multiple sclerosis, amyotrophic lateral sclerosis, epilepsy, concussion), conditions (cognitive impairment, learning disability), and normal function (activation of the human brain, sensitivity to pain). Specifically, our researchers are working on MRI methods sensitive to 1) tissue anisotropy, 2) tissue composition, 3) tissue metabolism, and 4) tissue function.

A sleep core/sleep reading center manager and the core director (Dr. Beth Malow) assist investigators with protocol development. It offers polysomnography, titrations with continuous positive airway pressure, multiple sleep latency testing, and wrist actigraphy (wireless devices that measure sleep-wake patterns non-intrusively based on accelerometers that measure activity vs. rest). The core provides for centralized scoring of polysomnography and actigraphy data for multicenter trials. In addition to the scoring component, emphasis is placed on the quality of the sleep recordings at the individual sites with close communication among the individual site technologists and the sleep core manager.
