It is a fee-for-service facility dedicated to providing proteomics support to the Carolinas HealthCare System, University of North Carolina at Charlotte, and other non-profit research organizations in the greater Charlotte region. The goal of the core is to facilitate proteomics analysis of clinical and biological samples by offering state-of-the-art instrumentation and proteomics expertise in preparing, analyzing, and bioinformatic interpretation of experimental data. The current mass spectrometry instrumentation includes an LTQ-XL and a TSQ Quantum Ultra, both with acquity UPLC systems, and an LTQ-XL Orbitrap-ETD with nano acquity UPLC. Additionally, we have six data analysis workstations.. Software platforms are used for proteomic data analysis include the Bioworks SEQUEST and Proteome Discoverer MASCOT search engines, Sieve and Scaffold Q+S for data compilation and quantification, PEAKS for de-novo sequencing, and the in-house script Intersect for use in comparison of more than two groups of samples. Pathway analysis is carried out using Metacore software. The Mass Spectrometry and Proteomics Core Facility uses high-throughput nano flow liquid-chromatography tandem mass spectrometry to identify proteins from one dimensional or two dimensional polyacrylamide gel slices. Sample preparation includes electrophoresis of the sample, gel staining and excision of regions of interest. The proteins are then digested within the gel slices using trypsin. The goal of the core is to facilitate proteomics analysis of clinical and biological samples by offering state-of-the-art instrumentation and proteomics expertise in preparing, analyzing, and bioinformatic interpretation of experimental data. The current mass spectrometry instrumentation includes an LTQ-XL and a TSQ Quantum Ultra, both with acquity UPLC systems, and an LTQ-XL Orbitrap-ETD with nano acquity UPLC. Additionally, we have six data analysis workstations.. Software platforms are used for proteomic data analysis include the Bioworks SEQUEST and Proteome Discoverer MASCOT search engines, Sieve and Scaffold Q+S for data compilation and quantification, PEAKS for de-novo sequencing, and the in-house script Intersect for use in comparison of more than two groups of samples. Pathway analysis is carried out using Metacore software. The Mass Spectrometry and Proteomics Core Facility uses high-throughput nano flow liquid-chromatography tandem mass spectrometry to identify proteins from one dimensional or two dimensional polyacrylamide gel slices. N108 Howell Science Complex | Greenville, NC 27858Welcome to the Genomics Core Facility, located in the Department of Biology at East Carolina University. We provide rapid, dependable and economical service for Sanger and next-gen sequencing to the research community of East Carolina University and researchers outside ECU. For Sanger sequencing, we have the 3130 Genetic Analyzer from Applied Biosystems, which delivers quality data of over 700 bases. The six color capability of the 3130 (with one color reserved for the size standard)allows for greater flexibility and cost-effectiveness in multiplexing samples for microsatellites, tRFLP or other fragment analysis methods. To determine the quality of RNA and DNA samples, we use the Agilent Bioanalyzer. The Bioanalyzer is sensitive enough to detect pico amounts of RNA, and the RIN (RNA Integrity Number), developed by Agilent, is the gold standard for RNA quality. Our newest service is next-gen sequencing, using the Ion Proton from Applied Biosystems. Using simple chemistry, the Proton is capable of sequencing whole genomes, transcriptomes and ChIP seq projects in just a few hours. Please use the links above to learn more about our services.

East Carolina University Flow Cytometry/Confocal Microscopy Core

115 Heart Drive, 4101 ECHI | Greenville, NC 27834This facility is a research supported resource which is part of the School of Medicine Biotechnology Program. Flow cytometry and Confocal microscopy are powerful techniques based on laser light excitation of fluorescent molecules. Flow cytometry is commonly used to study various aspects of cell function, including apoptosis, cell cycle, and immunophenotype analysis. The facility has two bench top flow analyzers: LSR II 4-laser 11-color flow cytometer and FACScan one laser 3-color cytometer (both Becton Dickinson). There is also a five-color cell sorter (BD FACSVantage) to isolate specific cell subsets from a complex mixture of cell types. Confocal microscopy is used primarily for cellular co-localization studies and three-dimensional reconstruction. The facility has a Zeiss 510 Laser-scanning Confocal Microscope which is capable of three-channel fluorescence imaging. Other instrumentation which is now part of the core facility are an iCys Laser Scanning Cytometer (Compucyte), which has similar capabilities as flow cytometry for samples on a slide or multiwell plate, and a PALM Laser Capture Microdissection system (Carl Zeiss), which allows excision and removal of specific target cells from a tissue or cell culture.

It is located on the third floor of the Howell Science Building in room N302. It is a recently renovated laboratory that serves as a teaching as well as research facility. The facility is used to train both undergraduate and graduate students. We are offering a graduate cell biology class every fall. In addition, we are teaching undergraduate workshops during the summer. These workshops are 2 weeks long and provide undergraduate students with necessary skills to work in a cell culture lab. The workshops are offered twice each summer. The facility contains enough equipment to accommodate up to eight students at a time. We have four biological safety cabinets, two CO2 incubators, two bench top centrifuges, a plate reader and plate washer, two inverted microscopes, and a compound microscope. Additionally, the facility also acts as a repository for cell line storage through the use of three cryo-storage units, which are available to all biology faculty members and their students.

About one-fifth of the space in the BRF (level one) is devoted to the Department of Entomology within CALS. Departments with research interests here are mostly within COS Biological Sciences. The facility was designed to house laboratory animals under conditions required by local and federal regulatory agencies. The BRF is regulated locally by the NC State University Institutional Animal Care and Use Committee (IACUC). The BRF is equipped with a backup electrical supply and air-conditioning system to insure the welfare of the animals. The BRF also operates a satellite facility on Centennial Campus. This facility support the research within Environmental Toxicology as well as provides space for College of Engineering, Biomedical Engineering. This Veterinarian provides weekly rounds and is available for animal health checks and research consults as requested. As a vertebrate developmental model, zebrafish offers several advantages over rodent models, such as small genome size, short generation time (embryogenesis), external development allowing embryo manipulation, and optical transparency of embryos. The large-scale production of zebrafish embryos is also a great advantage over rodent models, as orders of magnitude greater embryos can be produced from transgenic founders. The similarity in zebrafish and human genomes and conservation of gene function between zebrafish and human, combined with the experimental advantages of zebrafish, make zebrafish an extremely attractive model system for the development of human disease models and for high through-put chemical modifier (i.e., drug, natural product, and environmental toxicant) screening. Wild-type, mutant, and transgenic zebrafish lines are housed in tanks which recirculate water. The UNC CF/Pulmonary Research and Treatment Center Tissue Procurement and Cell Culture Core (TPC) was established to provide standardized cell cultures for CF research. The TPC serves as a central source of normal, CF and disease control cells, tissues and fluids for a wide array of uses. The TPC has prepared cells from approximately 6,900 human tissue specimens over the past 26 years, providing an average of 13 x 109 viable primary human airway epithelial cells per year over the past five years. Dr. Scott Randell, Core Director, and staff have extensive expertise in all aspects of tissue procurement and airway epithelial cell culture. The TPC is a national and international resource for donated tissues and share technology and reagents to help others develop equivalent tissue culture capabilities by sending out cells, tissues, and fluids to test key hypotheses related to current issues in CF pathogenesis and treatment. The latest advances in cell immortalization technology are used to create novel cell lines. An ever-increasing need for cells and services has broadened TPC functions to meet the growing research demands to study CF and other lung diseases.
