The Proteomics Shared Resource is a cooperative effort between the Siteman Cancer Center and the Washington University Department of Medicine. The facility is open to all Washington University investigators, with priority access given to cancer center members. The responsibilities of the Proteomics Shared Resource include: Providing global differential protein expression data on biological fluids, leukemias and solid tumors. Providing consultation on sample preparation and coordinating biostatistical-based experimental design. Archiving and retrieval of proteomics data for iterative analysis using protein informatics tools. Integration of proteomics data with other functional genomics data for cross queries through common data portals (in collaboration with Siteman’s Bioinformatics and Multiplexed Gene Analysis cores).

Room 7614 7th Floor 425 S. Euclid Avenue St. Louis, MO 63110Natural and therapeutically induced immune responses to cancer can prevent and/or facilitate cancer development/outgrowth. New cancer immunotherapies have shown remarkable capacities to induce durable responses in patients with advanced forms of cancer (such as stage IV melanoma) that are generally resistant to any other types of cancer therapy. These findings have significantly changed the thinking about cancer treatment and about using the immune system as an important and effective weapon against cancer. A centralized immune monitoring facility was launched in 2014 to facilitate translation of basic immune system research findings into the cancer immunotherapy arena, to support ongoing and new clinical cancer immunotherapy trials and to develop novel mechanistic insights into human immune system-cancer interactions. IML provides the means to monitor developing immune responses to new therapies in animal models of cancer treatment; to elucidate novel treatment strategies; or develop unique processes to evaluate treatment outcomes in translational studies. This shared resource provides the infrastructure and expertise to advance cancer immunotherapy outcomes.

Furthermore, the CBMI has extensive collaborations with WU centers and institutes and partnerships with global informatics initiatives and other academic centers in the region. Euclid Ave. Corner of Euclid and Forest Park Avenue St. Louis, MO 63110The Small-Animal Cancer Imaging (SACI) Shared Resource provides state-of-the-art facilities and infrastructure for MRI, PET, CT, Beta, SPECT, and optical imaging of mice, rats and other small laboratory animals that serve as models of cancer. Located in the heart of the Washington University Medical Center, SACI combines instrumental and intellectual capabilities found at few other institutions and serves a broad community of cancer scientists who have a pressing need for quantitative image analysis of small laboratory animal model systems. SACI provides access to and maintenance of small-animal MRI, PET, CT, and optical imaging scanners and ancillary facilities and routinely assists and trains researchers in imaging procedures and data analysis methods. In addition to supporting research applications of small-animal cancer imaging, SACI also provides research and development at the frontier of imaging technology in an effort to make the most powerful new imaging strategies available to its community of users. The ancillary small-animal research services and capabilities provided by SACI are extraordinary. Support resources within SACI include consultation for experiment planning and data analysis, animal-procedure assistance for maintenance and monitoring of physiologic status during imaging, and informatics support for local and remote data access, analysis, visualization, and archival.

Samples can be introduced by direct infusion or following separation using high performance liquid chromatography. Both qualitative and quantitative analyses are available by using full scan, single ion or selected reaction monitoring. A variety of tandem mass spectrometry experiments can be performed with ions produced by ESI or APCI methods. MU Variety Testing Program 4968 Rangeline Road South Columbia, MO 65201Selection of varieties that best fit a farmer's production goals and challenges is an essential part of profitable grain crop production. We provide the reliable, unbiased, up-to-date information that makes that selection possible. Each year we test over 600 corn, grain sorghum, and soybean varieties at 32 locations throughout Missouri. These 32 locations are distributed among four regions: North, Central, Southwest, and Southeast. The number of locations within a region depends on the specific test but varies from two to five. Companies enter their varieties into tests at one or more of these regions, but their entries must be placed at all locations within a region. The headquarters for the MU Variety Testing Program is the Bradford Research and Extension Center located in the heart of Missouri six miles east of Columbia. We also have personnel located at the Delta Research Center near Portageville, MO. The majority of our test locations are farmer fields and we appreciate the cooperation and dedication of our cooperators.

The core’s purpose is to make in vivo two-photon imaging and multidimensional data analysis widely available to WUSM researchers. The IVIC is operated as a recharge center to recover the cost of staff salaries, equipment and materials. The growing popularity of two-photon microscopy in the fields of cell biology, immunology and microbiology is in large part due to its unrivaled ability to image single-cell dynamics in native 3D tissue environments. Studying the behavior of individual cells is vital for understanding complex cellular systems, since many physiological processes are rare, asynchronous and tissue context dependent. With the appropriate fluorescence reporters, two-photon imaging can also serve as an in vivo assay for many aspects of cell activation and function, including motility, chemotaxis, antigen recognition, calcium influx, gene expression, proliferation, phagocytosis, cell-mediated cytotoxicity and apoptosis.

Washington University in St. Louis is home to one of the largest zebrafish facilities in the world. The facility is equipped with robotic feeding and cleaning systems; it is the world’s most modern. This facility allows large-scale, collaborative projects that would not be possible for individual investigators, helping scientists understand human development and disease, from birth defects and cancer to muscle and nerve disorders. The facility enables both large-scale forward genetic screens and generation of mutations in genes of interest, as well as transgenic strains. A common type of minnow, the zebrafish is popular in both scientific research and home aquariums. Zebrafish embryos are transparent and develop outside the body, making them useful for observing growth and development. The facility includes nearly 7,000 tanks, including a 2,000-tank nursery, and allows scientists to perform experiments requiring tens of thousands of fish. The robotic nature of the feeding process allows large numbers of fish to grow quickly facilitating genetic experiments. Beyond shear numbers of fish, the facility has other resources available to investigators. The facility also has the capabilities for obtaining, viewing, and manipulating fish embryos, including microinjections.

Washington University in St. Louis is home to one of the largest zebrafish facilities in the world. The facility is equipped with robotic feeding and cleaning systems; it is the world’s most modern. This facility allows large-scale, collaborative projects that would not be possible for individual investigators, helping scientists understand human development and disease, from birth defects and cancer to muscle and nerve disorders. The facility enables both large-scale forward genetic screens and generation of mutations in genes of interest, as well as transgenic strains. A common type of minnow, the zebrafish is popular in both scientific research and home aquariums. Zebrafish embryos are transparent and develop outside the body, making them useful for observing growth and development. The facility includes nearly 7,000 tanks, including a 2,000-tank nursery, and allows scientists to perform experiments requiring tens of thousands of fish. The robotic nature of the feeding process allows large numbers of fish to grow quickly facilitating genetic experiments. Beyond shear numbers of fish, the facility has other resources available to investigators. The facility also has the capabilities for obtaining, viewing, and manipulating fish embryos, including microinjections.

The Respiratory Viral Production Unit will provide standardized, fully-characterized respiratory viruses (HRV, HEV-D68, RSV, SeV and IAV) for use in human cells and the mouse model of acute infectious illness and post-viral asthma. 2). Animal Physiology Unit. The Mouse Physiology Unit houses instruments and personnel for invasive and non-invasive measurements of mouse lung function post-viral infection. Two BUXCO systems are currently on line within our laboratory space and will be utilized for lung physiology assessments. One system involves whole body plethysmography to assess airway reactivity through derivation of values for enhanced pause (Penh) following methacholine challenge (noninvasive), while the other involves invasive measurements of lung resistance (RL) and dynamic compliance (Cdyn) following methacholine challenge using the Buxco FinePoint system. 3) Drug Delivery/ Allergen-Challenge Unit. This Unit is experienced in conventional methods of drug delivery to animals, including oral, parenteral, and inhaled routes. Allergen-challenge with house dust mite to test for viral exacerbation is described below. 4) Cigarette-Smoke Exposure Unit. The Cigarette Smoking Unit has provided their facility and expertise services over 16 years.

Innovations in mouse transgenesis technologies include conditional gene modifications that inactivate gene function in tissue-specific manners, thereby overcoming embryonic lethality; and BAC transgenes to circumvent the position effects commonly found in standard transgenic mice. More recently, TALENs (TAL effector nucleases) and CRISPR/Cas9 systems have emerged as powerful new tools for genome modification in a variety of organisms and cell types, including mouse, rat, zebrafish, drosophila, C. elegans, and human stem cells and iPSCs (induced pluripotent stem cells). The Transgenic Vectors Core specializes in novel genome engineering technologies and vector construction using recombineering and isothermal assembly to generate complex gene modifications not possible with restriction enzymes. Large segments of genomic DNA such as those found on BACs (Bacterial Artificial Chromosomes) are now routinely modified, facilitating production of vectors that were previously impossible to generate The Transgenic Vectors Core designs and constructs TALENs, CRISPR/Cas9 and gene targeting vectors on a fee-for-service basis.
