It also performs enzyme assays to comprehensively diagnose a variety of metabolic disorders, such as galactosemia and many lysosomal storage disorders. Amino Acid Analysis & Organic Acid Analysis Amino acid analysis (blood, urine, CSF) and organic acid analysis (urine) are performed using state-of-the-art methodology and instrumentation. Enzyme Assays Enzyme assays for lysosomal storage diseases, galactosemia, galactokinase and other disorders are performed on peripheral blood leukocytes and cultured skin fibroblasts. We request that physicians check with the laboratory before requesting one of the analyses, in order to determine the type of specimen and quantity needed. Some of the tests must be scheduled in advance, and specimens cannot be collected on weekends or holidays. Tay-Sachs Carrier Testing Tay-Sachs carrier testing is performed on serum and peripheral blood leukocytes from adults (especially those with Ashkenazi Jewish ancestry) to determine if they carry the Tay-Sachs disease gene. Miscellaneous Tests Miscellaneous tests, including sugar chromatography, sialic acid quantitation and sulfites are available.

Equipment is available to process fixed tissue, embed in paraffin, section them and make routine hematoxylin and eosin staining as well as to perform immunohistochemistry and some special staining. Staff members are available for technical consultation regarding optimal harvesting and fixation of tissues, stains, histopathological procedures as well as for professional pathologic consultation and even imaging. Specimen Preparation For the best preservation of tissue morphology, the thickness of the tissue placed in fixative should be between 0.2 and 0.7 cm. Sections that are thicker require time for the fixative to permeate to the center, resulting in potential degradation. The volume of fixative should be at least 10 times that of the specimen. Fixation should be carried out as soon as possible after removal of the tissues, and for sufficient time to allow the permeation of the fixative into the tissue. Staff can provide the 10% buffered formalin by request. Tissues can remain in formalin for prolonged periods of time without compromising histology. However, after 24 hours, the longer the tissue is in formalin, the more antigenicity is lost.

The Fish Facility supports research projects from six research institute laboratories. In addition, investigators from Northwestern University (Chicago and Evanston campuses), DePaul University and University of Illinois at Chicago have used facility resources. The Fish Facility is outfitted with the Aquanee ring system for the rearing and maintenance of zebrafish. Four separate systems accommodate over 2,200 tanks for fry and adult fish, and provide optimal conditions for maintenance of up to 25,000 adult fish. All tanks are constantly supplied with water through inlet valves at a rate at which the water in the tanks is exchanged about three times per hour. The filtration system assures optimal water quality: tap water is purified by the Reverse Osmosis System, UV sterilization tower, particle prefilter, fine particle filter, and biofilter. Ten percent of fresh water is added to the system daily. Water quality (pH and conductivity) is monitored automatically. Water flow is also automatically controlled, and water level sensors with an alarm system are installed. The filter system requires minimal maintenance and sustains high water quality.
