The mission of the Flow Core is to provide sorting, analysis, experiment consultation, and instrument training. There are several instruments through out the EVC ranging from simple analog analyzers to digital sorters/analyzers. Analytical flow cytometry is used by the researcher to determine characteristics of subsets of cells or even single cells. Cell sorting is provided for both Bio-Safety level 2 and Bio-Safety level 3 samples. Sorting of cells is useful in the characterization of a particular subset of cells. These cells can then be manipulated in various ways by the investigator for a multitude of purposes.

Under contract by the National Institute of Allergy and Infectious Diseases, the Tetramer Core develops tetramer reagents that are not available from commercial sources. Tetramers are molecular complexes that can be engineered to bind only to specific types of immune cells – for instance, T-cells that respond to a particular disease-causing agent. Currently, the facility operates Bruker Avance III HD 600MHz, Bruker Avance III 400MHz, and Bruker Avance 400MHz spectrometers. The Bruker Avance III HD 600MHz NMR is equipped with triple resonance probe. It is dedicated to macromolecular research such as proteins, peptides, oligonucleotides, lipids, and carbohydrates. Both the Bruker AVIII 400MHz and AV 400MHz NMR spectrometers are equipped with BBFO probes where the broadband channel can observe frequencies from 109Ag to 19F. The two Bruker 400 MHz spectrometers are used primarily for the analysis of small molecules.

In the Coulter Multisizer, impedance is used to size and count cells as well as particles as they pass the through the orifice of an aperture. Flow Cytometry characterizes cell populations based on their light scatter and fluorescence properties. Using this information, cell subpopulations can be physically sorted into tubes, multi-well plates and on slides (See Sample Preparation Core).

The Facility can usually perform routine low-resolution analysis by EI, CI, ESI, nanoSpray, APCI and MALDI of small organic molecules and large biological molecules such as peptides, proteins, nucleic acids, lipids, oligosaccharides, polymers etc. The Facility also routinely conducts exact mass and elemental composition determination, tandem (MS/MS) experiments and GC and LC separations with MS detection as requested by researchers.

The repository collection includes a variety of specimen types, such as tumor tissue and cells, blood and other biofluids as well as normal specimens used as controls. The Biorepository also acts as a state-wide biorepository with samples being collected from 14 participating sites in the state of Georgia (the Bio-Repository Alliance of Georgia for Oncology or BRAG-Onc). The quality of all specimens are reviewed by a trained pathologist, and the information is captured in the tumor bank’s database. The tissue bank provides human specimens and laboratory services for basic and translational research to further the understanding of the cellular and molecular pathogenesis of human cancers.

Research applications include fracture healing, skeletal phenotyping, segmental bone defect repair, age-related effects on bone, joint degeneration, vascular remodeling, developmental biology, and microstructure of materials. The micro-CT lab is home to four micro-CT systems and a cluster of workstations to perform scanner operations, image reconstruction and quantitative analysis, and 3D visualization. There are two live animal scanners. The Scanco vivaCT 40, has a maximum field of view (FOV) of 38mm diameter and is capable of imaging live rodents and ex vivo specimens at 10-38um voxel resolutions. The Siemens Inveon has maximum FOV of 10cm and is capable of imaging live animals and specimens at 20-100um voxel resolutions. Additionally, there are two specimen only scanners. The Scanco uCT 40 has maximum FOV 36mm and is capable of voxel resolutions between 6 and 38um. The Scanco uCT 50 has maximum FOV 50mm and is capable of voxel resolutions between 0.5 and 50um. The power of these systems lies in the ability for users to extract quantitative morphometric and density data along with 3D models of their specimens. Some of the applications areas that have been impacted include assessing bone regeneration in large bone defects in vivo, quantifying trabecular bone microdamage, assessing bone repair in fracture healing models, and in vitro cell-mediated mineral growth on scaffolds. Though the systems were originally designed to measure hard tissues such as bone, the technology can also be used in conjunction with contrast agents in applications such as measuring degradative changes in articular cartilage and analyzing vascular remodeling after injury.

The Q-Scientifica SliceScope within the in vitro patch clamping rig is a compact upright microscope equipped with fully-motorized fixed stage, various electrode manipulators, a wide range of Olympus objectives and LED system (780 nm) to meet the demands of electrophysiology study. The electromagnetically shielded in vivo extracellular electrophysiology rig is constructed with various elements such as Zesis surgical microscopes, impressive 128-channel Tucker Davis Technologies data acquisition system (RZ2), Kopf stereotaxic frames and DC temperature controllers to enable stable, reliable and high-quality recordings. A complete LED driver system (Thorlabs) was equipped to this rig as well to facilitate optogenetic in vivo experiment. Automatic patch clamping devices (autopatchers) are also attached to both in vitro and in vivo patch clamping rigs to obtain high yield and high quality whole cell recordings. A mini surgery station, equipped with various types of stereotaxic systems, advanced anesthesia incubation chamber and isoflurane vaporizers, is also available for facilitating in vivo electrophysiology experiments.

For both basic and clinical investigators, the Analytical and Protein Profiling Laboratory will be a multifunctional resource providing analytical consultation, help in the preparation of manuscripts and proposals as well as sample analysis. This administrative structure ensures uniformity in investigator utilization, a common platform for marketing of services, thus promoting access, and increased utilization. Darien, GA 31305The University of Georgia Marine Institute (UGAMI), established in 1953, lies on a barrier island situated between the Atlantic Ocean and a pristine salt marsh-dominated estuarine ecosystem. The mission of the Institute is to support and conduct basic research on coastal processes involving the unique ecosystems of the Georgia coastline. UGAMI’s mission is also to provide access and facilities for graduate and undergraduate classes to experience field research and gain an appreciation of the Georgia coast. As a research facility literally embedded in the ecosystem of focus, UGAMI distinguishes itself with a long history of coastal research by resident and visiting researchers in a broad range of disciplines. As a result, significant contributions have been made to the understanding of the links between uplands and salt marsh estuaries to the coastal zone. UGAMI lies on Sapelo Island, a barrier island accessed from Meridian, Georgia by a passenger ferry which is run by the Georgia Department of Natural Resources 3 times per day (20 – 30 minutes between Meridian and Sapelo). UGAMI has a long-term lease for facilities totaling 1500 acres on the south end of Sapelo Island within and bordering the NOAA Sapelo Island National Estuarine Research Reserve (SINERR). The facility is very unique in its situation of relatively pristine surroundings, as well as the designation of the surrounding marshes as a NOAA SINERR (Fig. 1). This is a fortuitous and productive research situation. The remaining 2/3 of Sapelo Island are managed by the Georgia DNR as a Wildlife Management Area. There are approximately 100 full time residents on Sapelo Island, including UGAMI personnel, DNR personnel, and the small Hog Hammock community. Hog Hammock (500 acres) is the only privately owned property on Sapelo Island. Hog Hammock is a community of descendents of African-American plantation workers, and many members of the community are employed by UGAMI. UGAMI, Hog Hammock and the Georgia DNR have a long history of cooperation on Sapelo Island.

The research program includes diagnostics, prevention, and control strategies, prediction of disease outbreak, molecular epidemiology, and building a better understanding disease pathogenesis. SEPRL scientists also provides science-based solutions to exotic poultry diseases that inhibit international trade in poultry and poultry products. This especially includes developing practical and improved diagnostic tools including air samplers and creating vaccines. This includes diseases such as avian influenza, West Nile virus, Newcastle Disease and Marek's disease herpes virus. For example, SEPRL researchers have developed special tests to quickly detect avian influenza and Newcastle disease outbreaks as well as biosecurity measures to protect the poultry industry from these diseases. These tests have been instrumental in protecting the U.S. poultry industry from disease issues that could impact international trade. They also invented a highly sensitive diagnostic test to detect viruses associated with poult enteritis complex, The test allows the detection of several types of viruses at one time. Current diagnostic methods for PEC-associated viruses have limitations because of poor analytic specificity and sensitivity.
